This study explores how a new medicine called **NMRA-335140** affects people with **Major Depressive Disorder (MDD)**. MDD is a condition that makes people feel very sad and hopeless for a long time. The study is **randomized** (participants are randomly assigned to treatment groups), **double-blind** (neither participants nor researchers know who receives the actual drug or a placebo), and **placebo-controlled** (some participants receive a placebo, a harmless pill that has no effect). It lasts for about **6 weeks**. Participants will take either the study medicine or a placebo. At the end, those who finish the study can join a longer study if they agree and qualify.
- **Duration**: Up to 35 days for screening plus 6 weeks of treatment.
- **Participation**: Must have MDD without psychotic features and meet certain health criteria.
- **Exclusion**: Cannot have certain other mental health issues, substance abuse, or be actively suicidal.
